Thermo Scientific SMART Digest Kit
Obtain high quality analytical results from protein digests by using Thermo Scientific™ SMART Digest™ Kits. A significant advance in sample preparation for biopharmaceutical and protein research, the kits provide fast and simple protein digestion with high reproducibility, high sensitivity, and high levels of data quality in a format that's compatible with automation. The SMART Digest Kits are a significant improvement over current in-solution protein digestion technologies, which are not reproducible, have poor sensitivity, and employ protracted methodologies not amenable to automation.]

High-precision, Automated Peptide Mapping of Proteins
Hear Dr. Amy Farrell of the National Institute of Bioprocessing Research and Training (NIBRT) discuss the development of a robust and reproducible, high-precision, automated, protein digestion workflow that is appropriate as an easy-to-use, general approach to peptide mapping characterization with both LC-only and LC-MS processes. The quality of peptide maps was confirmed with multiple example proteins and high sequence coverage was possible using high-resolution, accurate-mass mass spectrometry.

The aim of this work was to demonstrate the use of the new Thermo Scientific™ Vanquish™ Duo UHPLC system and enable tandem analysis with two columns in parallel, addressing productivity and throughput improvement of existing LC-MS methods.
To compare the results achieved by using the newly developed Thermo Scientific™ SMART Digest™ kit to those obtained from classic in-solution protein digestion methods, focusing on protein sequence coverage and identified post-translational modifications (PTMs), including deamidation, oxidation, and glycosylation.
To perform complete characterization of the lysine-linked antibody drug conjugate (ADC) trastuzumab emtansine using both intact mass and peptide mapping approaches with a single benchtop mass spectrometer. Critical quality attribute (CQA) measurements include drug-to-antibody ratio (DAR) and glycosylation pattern via intact mass analysis, and conjugation site localization via peptide mapping.
Peptide mapping is a core analytic in biotherapeutic development. Yet it is slow, involves painstaking sample preparation, and is prone to errors. This booklet will guide you through the rough terrain of peptide mapping and upgrade you with a solution that’s better, wherever on the map you are.
